基因组的设计与合成可显著扩大序列改造的规模,有望突破现有的基因组功能认知,拓展人工生命体的应用。课题组负责人前期参与了世界首个真核生物基因组合成计划,构建了迄今为止最长的真核生物线性合成染色体,首次鉴定了组蛋白2-hydroxyisobutyrylation的修饰酶和去修饰酶,开创的MRA 层级组装技术和ReSCuES筛选技术解决了合成染色体设计、构建和应用等方面的系列难点,成果发表在Science(封面专刊)、Cell Res、Nat Commun和Genome Biol等期刊上。研究成果获得深圳市自然科学一等奖,入选2017年度中国高等学校十大科技进展和中国科学十大进展。本课题组未来将继续致力于基因组相关的基础理论研究、关键技术突破和新型应用开发。
Synthetic genomics is an important part of synthetic biology, which is expected to innovate our understanding about genome function and to significantly improve the depth and ability of genome engineering. We participated in the world's first eukaryotic genome synthesis project, constructed the longest eukaryotic linear synthetic chromosome so far, identified the Writers and Erasers of histone 2-hydroxyisobutyrylation, and created technologies for synthetic chromosome design, construction and application. Our results have been published in journals such as Science (cover story and special issue), Cell Res, Nat Commun and Genome Biol. In the future, our research group will continue to focus on the basic theories, key technologies and new applications related to genome.
